I have developed a painful lump between the first and second knuckles on the back of my right hand. It is very painful and pronounced when I make a fist.

Hi Linda - it’s most likely a ganglion cyst.
They are really common between joints, especially in the hand and wrists. It’s a fluid filled sac that can get bigger and smaller.
I don’t think it’s anything to worry about and best left alone.
